Protein conformational disorder and enzyme catalysis.

Though lacking a well-defined three-dimensional structure, intrinsically unstructured proteins are ubiquitous in nature. These molecules play crucial roles in many cellular processes, especially signaling and regulation. Surprisingly, even enzyme catalysis can tolerate substantial disorder. This observation contravenes conventional wisdom but is relevant to an understanding of how protein dynam...

PPIase catalysis by human FK506-binding protein proceeds through a conformational twist mechanism.

FK506-binding protein (FKBP) catalyzes the cis-trans isomerization of the peptidyl-prolyl amide bond (the PPIase reaction) and is the major intracellular receptor for the immunosuppressive drugs FK506 and rapamycin. One mechanism proposed for catalysis of the PPIase reaction requires attack of an enzyme nucleophile on the carbonyl carbon of the isomerized peptide bond. An alternative mechanism ...

Catalysis by enzyme conformational change as illustrated by orotidine 5'-monophosphate decarboxylase.

An energy decomposition scheme has been used to elucidate the importance of the changes of enzyme conformational substates to the reduction of the activation barrier in enzyme-catalyzed reactions. This analysis may be illustrated by the reaction of orotidine 5'-monophosphate decarboxylase, which exhibits a remarkable rate enhancement of over 17 orders of magnitude compared to the uncatalyzed pr...

Microwave activation of enzymatic catalysis.

Microwave irradiation can be used to regulate biocatalysis. Herein, the utilization of hyperthermophilic enzymes in a microwave reactor is reported. While these enzymes are inactive at low temperatures, they can be activated with microwave irradiation. To the best of our knowledge, this is the first illustration of a specific microwave effect in enzymatic catalysis.

Binding Energy and Enzymatic Catalysis

For nearly 100 years, the extraordinary catalytic power of enzymes has fascinated biochemists (1). In a recent article in this Journal, Splittgerber (2) analyzed several ofthe factors by which enzymes are thought to effect catalysis, including proximity and orientation effects, substrate strain, acidbase catalysis, and nucleophilic catalysis. Here we will discuss the fundamental role that the f...
